No, they didn't.
We have no idea if Horemheb had any kind of link to the throne. For years he has been associated with Paatenemheb, a highly-placed noble at Akhetaten, who was also the top military man. I personally think it highly unlikely that two men with similar names & titles would be around, especially since Paatenemheb disappears & Horemheb arrives.
We also don't know what the relationships between Ankhesenamun, Horemheb & Aye were like. Aye had at best a tangental connection to the throne. Horemheb's claim may have been much better.
